Great idea!
Some things I missed:
* better feedback for what is going on (what phase this is, what can I do right now) * more enemies/action?
To make the UI easier to parse I think you could add some panels for the various sections, along with a some "tutorial" type pointers on what to do (e.g. when the towers can be placed, show a big arrow pointing at them, then when a tower is picked up highlight somehow the tiles where the tower can be placed).
I think the mechanic would be easier/faster to explore in a 2d game.
